Serum Formulation: Ingredient Compatibility, Stability, and Scaling Pitfalls

For skincare brands, serums can become an undisputed powerhouse. However, translating serum from a marketing concept to a stable, scalable manufactured product requires more than simply choosing popular ingredients.

If your brand is looking to develop custom serums, understanding the chemistry behind formulation and the realities of serum manufacturing is the first step toward a successful, scalable product launch.

Let’s look at four in-demand actives: Vitamin C, Niacinamide (Vitamin B3), Hyaluronic Acid, and Squalene, and how they’re used in modern formulations.

The Science of Scale: An Expert’s Perspective

As premier serum manufacturers, FP Labs understands that combining high concentrations of active ingredients introduces complex formulation challenges, strict manufacturing restrictions, and crucial packaging considerations. Success ful serum development depends on formulation design, delivery system, processing conditions, stability and packaging.

To build a serum that truly performs, you have to look past the ingredient list and understand how molecules will behave at scale.

“Anyone can combine ingredients in a laboratory beaker. The real science begins with understanding how those ingredients interact throughout formulation, manufacturing, and shelf life. Every serum requires a deep understanding of ingredient compatibility, delivery technology, rheology, processing conditions, manufacturing scale-up, and packaging interactions. At FP Labs, we engineer the formulation, the manufacturing process, and the package as a single integrated system, ensuring every serum delivers exceptional stability, elegant aesthetics, and consistent performance—from the first laboratory batch to full-scale commercial production.” – Paul Marotta, Sr. Director R&D Formulation and Process Engineering

Core Actives: Manufacturing Restrictions and Considerations

Vitamin C (L-Ascorbic Acid vs. Derivatives)

  • Why Consumers Love It. Brightens skin, reduces the appearance of dark spots, promotes collagen production, protects skin from environmental stressors, neutralizes free radicals.
  • The Manufacturing Reality. Vitamin C is notoriously unstable. L-ascorbic acid degrades rapidly when exposed to heat, light, and oxygen. During manufacturing, specialized handling is required, sometimes necessitating nitrogen-purged tanks to prevent oxidation during blending. We often advise brands to use stabilized derivatives such as Sodium Ascorbyl Phosphate. This helps ensure shelf stability and works well with higher-pH formulas.
  • Packaging Consideration. Packaging is critical for Vitamin C to retain its stability. Airtight packaging is strongly recommended over traditional droppers to limit oxygen exposure. Amber colored bottles also tend to be used because the brown glass filters out rays from the sun and slows the breakdown of the active.

Niacinamide (Vitamin B3)

  • Why Consumers Love It. Helps regulate sebum production (good for acne-prone skin), minimizes redness and blotchiness, improves skin barrier function, reduces moisture loss and the appearance of fine lines and enlarged pores.
  • The Manufacturing Reality: While versatile (it works across a wide pH range of 5-7), Niacinamide is highly pH-dependent. If combined with anything highly acidic, such as pure Vitamin C or AHAs, in a formulation that drops below pH 5.0, Niacinamide can hydrolyze to nicotinic acid, which can cause severe skin flushing.

When developing custom serums, our chemists carefully buffer the pH to safely blend multi-active formulas without compromising efficacy or causing consumer irritation.

    Hyaluronic Acid: The Ultimate Hydrator

    • Why Consumers Love It. Extreme hydration, instant skin plumping, and improved skin elasticity with consistent use.
    • The Manufacturing Reality. Hyaluronic Acid (HA) functions well across a wide pH range and is generally non-irritating. The challenge in commercial scale-up is viscosity and shear sensitivity. High-molecular-weight HA acts as a thickener, which can cause extreme shear stress during the blending and filling process.

Formulators have to balance molecular weights to achieve the desired skin penetration while ensuring the bulk product still flows efficiently through industrial filling lines.

    Squalene: The Lightweight Oil

    • Why Consumers Love It. Provides lightweight, non-comedogenic moisture, mimics the skin’s natural lipids, reinforces the skin barrier, and protects against free radical damage.
    • The Manufacturing Reality. Highly stable and compatible with most actives. As an emollient lipid, squalene in a water-heavy serum requires a highly stable emulsion system. Inadequate emulsification leads to phase separation over time.

Our Research & Development team uses advanced homogenizers and thermal testing to ensure squalene-based serums maintain a uniform, elegant texture throughout their shelf life.

    How to Combine Actives Safely

The art of serum formulation isn’t just about choosing powerful actives; it’s about making sure they work together harmoniously. The best active ingredient delivers little value if it never reaches its intended destination within the skin. Designing the appropriate delivery system is just as important as selecting the active itself. Our scientists optimize penetration, maximize bioavailability, minimize degradation, and maintain ingredient stability throughout the product’s shelf life.

    • pH Compatibility – Vitamin C in its purest form requires a low pH, which can destabilize niacinamide or cause irritation. Stabilized forms of Vitamin C are often better for multi-active serums.
    • Stability Considerations – Antioxidants like Vitamin E can help stabilize Vitamin C, extending shelf life.
    • Irritation Risk – Combining strong actives (like high-dose Vitamin C + niacinamide) can increase sensitivity. Using lower concentrations or encapsulated options reduces this risk.
    • Texture Balance – A mix of humectants that draw moisture into the skin (like hyaluronic acid), emollients that soften, smooth, and lubricate the skin’s surface (like squalene), and water-soluble actives that help the serum feel elegant and deliver results.

    Going From Bench to Bulk

Formulating custom serums in a lab is only half the battle. Scaling a formula from a 100mL lab beaker to a 1,000-gallon compounding tank changes the mix’s thermodynamic properties.

By partnering with an experienced manufacturer like FP Labs, brands can ensure that the manufacturing process is meticulously documented and executed to deliver identical, high-quality results, batch after batch. When it comes to scaling a formula, it’s all about consistency.

    Serum Manufacturing FAQs

What are the primary restrictions when manufacturing custom serums?

The most common restrictions involve ingredient stability, thermal sensitivity, and pH compatibility. Some actives simply cannot coexist in a single base without neutralizing each other or causing rapid oxidation.

Additionally, light- and oxygen-sensitive actives restrict both the atmospheric manufacturing environment and the final packaging formats. As expert serum formulators, we’re well-versed in all these restrictions and the best ways for your brand to navigate them!
